Summary of Meeting:
Although the role of hypercholesterolemia in the etiology of atherosclerosis is now established beyond doubt, understanding the complex molecular and cellular events involved in the artery wall are a prerequisite for an improved understanding of the pathogenesis of atherosclerosis, and for designing specific and improved interventions to prevent clinical disease. The atherosclerotic lesion has many properties of a chronic inflammatory disease, involving activated monocyte/macrophages, endothelial cells and smooth muscle cells, as well as humoral and cell-mediated immunological responses. Oxidative events play a central pathogenic role, both in the generation of modified lipids and proteins that mediate proinflammatory events, as well as in the formation of foam cells. Molecular events involved in regulation of key cellular events, including the role of nuclear receptors, that control cell trafficking into the artery, expression of cytokines, growth factors, metalloproteinases, and tissue factor, as well as regulation of cellular growth, migration, apoptosis and death are beginning to be addressed. In addition, recent experimental evidence indicates an important pathogenic role for angiogenesis. The proposed program will be held jointly with a Symposium dedicated to the role of angiogenesis and vascular biology, and should broaden the appeal of this meeting to investigators interested in these two interrelated topics. Specific sessions will be devoted to the role of oxidative stress, immunological mechanisms, macrophage biology, endothelial cell biology and scavenger receptors, as they apply to the pathogenesis of atherosclerosis. Joint plenary sessions will deal at a more fundamental level with nuclear receptors, cell trafficking and apoptosis/angiogenesis. This Keystone Symposium is intended to foster communication between investigators involved in understanding various mechanisms involved in vascular biology and atherosclerosis, and that new insights will be gained that will lead to novel and effective interventions to prevent this disease.
