Meeting Summary

Cancer immunotherapy has transformed cancer treatment, yet durable responses remain limited to subsets of patients and tumor types. Resistance, inadequate immune priming, antigen loss, hostile tumor microenvironments, treatment-associated toxicities, and barriers to clinical translation continue to constrain the impact of current approaches. Key knowledge gaps include understanding why promising therapeutic targets failed during translation and how emerging tools and modalities move the field beyond established checkpoint-based paradigms.

This Keystone Symposia conference on Cancer Immunotherapy: New Pathways and Innovating Therapeutics aims to define the next generation of cancer immunotherapy. The program examines both successful and unsuccessful therapeutic strategies, emphasizing actionable lessons from trial design, strategies, manufacturing, and immune-related adverse events. Plenary sessions cover engineered cell therapies, bispecific and trispecific engagers, advanced cancer vaccines, novel antigen spaces, myeloid-directed therapies, AI-enabled discovery, and strategies to overcome immune escape.

The conference brings together immunologists, physician-scientists, oncologists, technologists, and industry leaders. Mechanistic insight connects with therapeutic design by integrating drug development with basic tumor immunology, cancer biology, clinical investigation, and new computational approaches, allowing attendees to identify successful and failed translation and to define more effective paths from discovery to patients.

Rather than centering on established checkpoint therapies, this conference emphasizes emerging approaches and the unresolved barriers determining success or failure. Candid debates and speakers address failures, toxicities, and bottlenecks alongside new opportunities. The conference is particularly timely as these emerging modalities rapidly reshape the field, offering attendees rare insights.
